The HMB-45 antibody is a monoclonal antibody that targets melanosomes, specifically recognizing gp100, a protein located in the melanosomes of melanocytes. Melanosomes are organelles responsible for the synthesis and storage of melanin, the pigment responsible for skin color. The [HMB-45] antibody is widely used in immunohistochemistry (IHC) to identify melanocytic lesions, such as melanomas, and to assess melanocytic differentiation. It is particularly useful in distinguishing malignant melanomas from other non-melanocytic tumors because HMB-45 is strongly expressed in melanocytes and melanomas but typically absent in benign lesions like nevi. This makes it a valuable tool for diagnosing cutaneous melanoma and evaluating melanocytic tumors in clinical pathology, helping to differentiate malignant melanocytic growths from other skin lesions.
